Cap-Girardeau (en anglais : Cape Girardeau) est une ville du Missouri, dans le Comté de Cape Girardeau aux États-Unis.
Cap-Girardeau possède un aéroport (Cape Girardeau Municipal Airport, code AITA : CGI).
Le nom de la ville vient de Jean Baptiste de Girardot, qui créa un poste de traite provisoire à cet endroit vers 1733; C'était un militaire français basé à Kaskaskia (actuel Illinois). Le Cap était un promontoire rocheux qui dominait le Mississippi, promontoire qui fut détruit lors de la construction du chemin de fer.
