Les protothériens (Prototheria) sont un groupe de mammifères, une sous-classe, qui présentent des caractères qui les rapprochent des reptiles. D'après certains auteurs, ils constituent l'ordre actuel des monotrèmes et des ordres éteints tel que Docodonta, Eotheria, selon d'autres auteurs ce taxon doit être remplacé par celui des Australosphenida^[1]. Une autre controverse concerne la position relative de ce taxon vis à vis des Metatheria. Certains indices morphologiques font penser que les Metatheria sont proches des Placentaires alors que les monotrèmes ont évolué séparément depuis plus longtemps. Cependant des analyses récentes portant sur l'ADN mitochondrial semblent indiquer que les monotrèmes et les marsupiaux forment un clade, sans les placentaires. Les analyses employant de grands segments d'ADN confirment la vue classique.

Taxonomie

Le terme de prototheria dérive du grec prōtos qui signifie premier et de thēr qui signifie animal sauvage. Ce terme peut donc se traduire par premier animal sauvage. Il a été proposé par Thomas Henry Huxley.
