La rotoscopie est, à l'origine, une technique d'animation crée par Dave et Max Fleischer aux alentours de 1914, consistant à transformer une scène filmée en un dessin animé. Elle permet d'obtenir des animations plus fluides, au rendu naturel. Ce procédé est notamment utilisé dans le dessin animé du Seigneur des Anneaux réalisé par Ralph Bakshi.

Les rotoscopes traditionnels utilisaient une table de projection transparente, sur laquelle apparaissait chaque image du film. Le dessinateur pouvait alors suivre les contours des formes réellement filmées, et les redessiner sur un calque approprié.

De nos jours, avec l'utilisation des outils informatiques graphiques, le terme rotoscopie signifie la retouche ou le découpage manuel, image par image, d'un film numérisé, afin d'en supprimer certains éléments ou d'en ajouter d'autres, notamment de décor.

Par exemple, pour un film historique, un plan est tourné avec des acteurs costumés, dans un paysage naturel. On souhaite supprimer des éléments anachroniques apparaissant à un endroit, ou même changer tout un arrière-plan pour des raisons esthétiques. À l'aide de logiciels appropriés (mais il s'agit d'un travail manuel) des infographistes modifient le film initial, en ne conservant que ce que le réalisateur souhaite. On peut alors y incorporer d'autres éléments (naturels ou générés par ordinateur).
